It plays an essential role in maintenance of normal retinal and cochlear function. Mutations in this gene result in hearing loss and Usher Syndrome Type IF (USH1F). Extensive alternative splicing resulting in multiple isoforms has been observed in the mouse ortholog. p.Asp1596Glu in exon 33 of PCDH15: This variant is not expected to have clinical significance due to a lack of conservation across species, including mammals. O f note, 7 mammals (pika, dog, ferret, panda, Pacific walrus, Weddell seal, eleph ant) have a glutamic acid (Glu) at this position despite high nearby amino acid conservation. This variant has also been identified in 2/10406 African chromoso mes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; d bSNP rs200659780).
