Genes affected
PCDH15 (HGNC:14674): (protocadherin related 15) This gene is a member of the cadherin superfamily. Family members encode integral membrane proteins that mediate calcium-dependent cell-cell adhesion. It plays an essential role in maintenance of normal retinal and cochlear function. Mutations in this gene result in hearing loss and Usher Syndrome Type IF (USH1F). Extensive alternative splicing resulting in multiple isoforms has been observed in the mouse ortholog. Similar alternatively spliced transcripts are inferred to occur in human, and additional variants are likely to occur. [provided by RefSeq, Dec 2008]

Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsApr 21, 2022The c.4788T>G (p.D1596E) alteration is located in exon 33 (coding exon 32) of the PCDH15 gene. This alteration results from a T to G substitution at nucleotide position 4788, causing the aspartic acid (D) at amino acid position 1596 to be replaced by a glutamic acid (E). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Likely benign, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineMar 07, 2017p.Asp1596Glu in exon 33 of PCDH15: This variant is not expected to have clinical significance due to a lack of conservation across species, including mammals. O f note, 7 mammals (pika, dog, ferret, panda, Pacific walrus, Weddell seal, eleph ant) have a glutamic acid (Glu) at this position despite high nearby amino acid conservation. This variant has also been identified in 2/10406 African chromoso mes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; d bSNP rs200659780). -
