In summary, this variant is a rare missense change with uncertain impact on protein function. It has been reported in both the population and affected individuals, but the available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change leads to channel mediated currents with accelerated inactivation, but the clinical implications of this finding are uncertain (PMID: 12185453). This variant has been reported in two individuals affected with long QT syndrome (PMID: 12185453, 24606995). ClinVar contains an entry for this variant (Variation ID: 67611). This variant is present in population databases (rs199473364, ExAC 0.006%). This sequence change replaces valine with methionine at codon 65 of the KCNE2 protein (p.Val65Met). The valine residue is highly conserved and there is a small physicochemical difference between valine and methionine. -
