This variant was determined to be of uncertain significance according to ACMG Guidelines, 2015 [PMID:25741868]. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.I1127S variant (also known as c.3380T>G), located in coding exon 20 of the DICER1 gene, results from a T to G substitution at nucleotide position 3380. The isoleucine at codon 1127 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
