Genes affected
DICER1 (HGNC:17098): (dicer 1, ribonuclease III) This gene encodes a protein possessing an RNA helicase motif containing a DEXH box in its amino terminus and an RNA motif in the carboxy terminus. The encoded protein functions as a ribonuclease and is required by the RNA interference and small temporal RNA (stRNA) pathways to produce the active small RNA component that represses gene expression. This protein also acts as a strong antiviral agent with activity against RNA viruses, including the Zika and SARS-CoV-2 viruses. Alternative splicing results in multiple transcript variants. ACMG classification

Classification made for transcript

Verdict is Likely_benign. Variant got -2 ACMG points.

PP2
Missense variant where missense usually causes diseases, DICER1
BP4
Computational evidence support a benign effect (MetaRNN=0.07304475).
BP6
Variant 14-95104016-A-C is Benign according to our data. Variant chr14-95104016-A-C is described in ClinVar as [Conflicting_classifications_of_pathogenicity]. Clinvar id is 412046.We mark this variant Likely_benign, oryginal submissions are: {Uncertain_significance=2, Likely_benign=1}.

Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsApr 06, 2023The p.I1127S variant (also known as c.3380T>G), located in coding exon 20 of the DICER1 gene, results from a T to G substitution at nucleotide position 3380. The isoleucine at codon 1127 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
