Genes affected
PIK3R1 (HGNC:8979): (phosphoinositide-3-kinase regulatory subunit 1) Phosphatidylinositol 3-kinase phosphorylates the inositol ring of phosphatidylinositol at the 3-prime position. The enzyme comprises a 110 kD catalytic subunit and a regulatory subunit of either 85, 55, or 50 kD. This gene encodes the 85 kD regulatory subunit. Phosphatidylinositol 3-kinase plays an important role in the metabolic actions of insulin, and a mutation in this gene has been associated with insulin resistance. Alternative splicing of this gene results in four transcript variants encoding different isoforms. [provided by RefSeq, Jun 2011]

ACMG classification

Classification was made for transcript

Our verdict: Benign. The variant received -11 ACMG points.

BP4
Computational evidence support a benign effect (MetaRNN=0.09048909).
BP6
Variant 5-68280542-G-A is Benign according to our data. Variant chr5-68280542-G-A is described in ClinVar as Conflicting_classifications_of_pathogenicity. ClinVar VariationId is 576303.
BS1
Variant frequency is greater than expected in population sas. GnomAdExome4 allele frequency = 0.0000486 (70/1441706) while in subpopulation SAS AF = 0.000595 (51/85696). AF 95% confidence interval is 0.000465. There are 4 homozygotes in GnomAdExome4. There are 54 alleles in the male GnomAdExome4 subpopulation. Median coverage is 30. This position passed quality control check.
BS2
High Homozygotes in GnomAdExome4 at 4 AD,AR gene
