The protein encoded by this gene serves as the small subunit of this complex and is a member of the adaptin protein family. To our knowledge, no occurrence of c.319delG in individuals affected with Pettigrew Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Downstream variants (c.367C>T/p.Q123* and c.426+1G>T) were found in HGMD and classified in ClinVar as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
