Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect DMD function (PMID: 14962982). This variant disrupts the p.Cys3313 amino acid residue in DMD. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 12632325, 24302611, 28859693; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. ClinVar contains an entry for this variant (Variation ID: 287224). This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 3313 of the DMD protein (p.Cys3313Tyr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of DMD-related conditions (PMID: 31443951, 33644936). -
