This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 96 of the SDHB protein (p.Gly96Asp). This variant is present in population databases (rs778952116, gnomAD 0.02%). This missense change has been observed in individuals with paraganglioma, pheochromocytoma or renal cell carcinoma (PMID: 16317055, 17538171, 19075037, 28891197; internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 428916). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic. -

The p.G96D pathogenic mutation (also known as c.287G>A) is located in coding exon 4 of the SDHB gene. The glycine at codon 96 is replaced by aspartic acid, an amino acid with similar properties. This variant has been reported in multiple individuals diagnosed with paragangliomas and/or pheochromocytomas (Benn DE et al. J Clin Endocrinol Metab. 2006 Mar;91(3):827-36; Timmers HJ et al. J Clin Endocrinol Metab. 2008 Dec;93(12):4826-32; Ghayee HK et al. Endocr. Relat. Cancer, 2009 Mar;16:291-9; Jochmanova I et al. J. Cancer Res. Clin. Oncol., 2017 Aug;143:1421-1435; Samuel N et al.
