Variant summary: LAMB3 c.2036C>T (p.Pro679Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8.8e-05 in 251406 control chromosomes (gnomAD). This frequency is not significantly higher than expected for a pathogenic variant in LAMB3 causing Junctional Epidermolysis Bullosa (8.8e-05 vs 0.0024), allowing no conclusion about variant significance. c.2036C>T has been reported in the literature in individuals affected with Junctional Epidermolysis Bullosa, non-Herlizt type (Pulkkinen_1995, Al Zahrani_2019). These reports do not provide unequivocal conclusions about association of the variant with Junctional Epidermolysis Bullosa. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
