This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 750 of the RET protein (p.Ala750Gly). This variant is present in population databases (rs752830000, gnomAD 0.004%). This missense change has been observed in individual(s) with breast cancer (PMID: 28202063). ClinVar contains an entry for this variant (Variation ID: 560866). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.A750G variant (also known as c.2249C>G), located in coding exon 12 of the RET gene, results from a C to G substitution at nucleotide position 2249. The alanine at codon 750 is replaced by glycine, an amino acid with similar properties. This alteration has been reported in an individual from a Macedonian cohort of unselected probands with medullary thyroid cancer (Jovanovic R et al. Pril (Makedon Akad Nauk Umet Odd Med Nauki) 2015 ;36(1):93-107). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
