This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder. [provided by RefSeq, Aug 2010]

The proline at codon 424 is replaced by histidine, an amino acid with similar properties. This alteration has been reported in individuals with breast cancer, including male breast cancer (Tung N et al. Cancer, 2015 Jan;121:25-33; Penkert J et al. Breast Cancer Res, 2018 Aug;20:87; Rizzolo P et al. Int J Cancer, 2019 Jul;145:390-400). This alteration was also detected in both the germline and the tumor in a female diagnosed with ovarian cancer (Coombs CC et al. Clin. Cancer Res., 2018 Jun). This variant was reported in 1/5560 prostate cancer cases and in 0/3353 controls of European ancestry (Karlsson Q et al. Eur Urol Oncol, 2021 Aug;4:570-579). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. These reports therefore, do not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance.
