The HBB: c.91A>G; p.Arg31Gly, also known as Arg30Gly when numbered from the mature protein, rs35684407, HbVar ID: 815) is reported heterozygous in the literature in multiple individuals affected with beta(0) thalassemia trait (See HbVar database and references therein). This variant is absent from the Genome Aggregation Database (v2.1.1), indicating it is not a common polymorphism. Computational analyses (Alamut Visual Plus v.1.5.1) predict that this variant may impact splicing by weakening the nearby canonical donor splice site. Additionally, another variant at this codon (Hb Monroe variant, c.92G>C; p.Arg31Thr, HbVar ID: 290) has been reported in individuals with beta (0) thalassemia, is shown to affect splicing, and is considered pathogenic (Vidaud 1989).
