Genes affected
POLE (HGNC:9177): (DNA polymerase epsilon, catalytic subunit) This gene encodes the catalytic subunit of DNA polymerase epsilon. The enzyme is involved in DNA repair and chromosomal DNA replication. Mutations in this gene have been associated with colorectal cancer 12 and facial dysmorphism, immunodeficiency, livedo, and short stature. [provided by RefSeq, Sep 2013]

Submissions by phenotype

Likely benign, criteria provided, single submitterclinical testingGeneDxOct 25, 2017This variant is considered likely benign or benign based on one or more of the following criteria: it is a conservative change, it occurs at a poorly conserved position in the protein, it is predicted to be benign by multiple in silico algorithms, and/or has population frequency not consistent with disease. -

Familial ovarian cancer Benign:1
Likely benign, no assertion criteria providedclinical testingDepartment of Pathology and Laboratory Medicine, Sinai Health System-The POLE c.3378+7G>T variant was not identified in the literature. The variant was identified in dbSNP (rs755370377) as “with other allele” and ClinVar (interpreted as "benign" by Invitae and Quest Diagnostics and "likely benign" by GeneDx and Counsyl). The variant was identified in control databases in 30 of 277,184 chromosomes at a frequency of 0.0001 increasing the likelihood this could be a low frequency benign variant (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: European in 4 of 126,694 chromosomes (freq: 0.00003), Ashkenazi Jewish in 25 of 10,150 chromosomes (freq: 0.002), and South Asian in 1 of 30,778 chromosomes (freq: 0.00003). The variant was not observed in the African, Other, Latino, East Asian and Finnish populations. The variant occurs at a non-conserved nucleotide outside of the consensus splicing sequence and in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) did not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time although we would lean towards a more benign role for this variant. This variant is classified as likely benign. -
