The variant was identified in dbSNP (rs755370377) as “with other allele” and ClinVar (interpreted as "benign" by Invitae and Quest Diagnostics and "likely benign" by GeneDx and Counsyl). The variant was identified in control databases in 30 of 277,184 chromosomes at a frequency of 0.0001 increasing the likelihood this could be a low frequency benign variant (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: European in 4 of 126,694 chromosomes (freq: 0.00003), Ashkenazi Jewish in 25 of 10,150 chromosomes (freq: 0.002), and South Asian in 1 of 30,778 chromosomes (freq: 0.00003). The variant was not observed in the African, Other, Latino, East Asian and Finnish populations. The variant occurs at a non-conserved nucleotide outside of the consensus splicing sequence and in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) did not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time although we would lean towards a more benign role for this variant. This variant is classified as likely benign. -
