The encoded protein is part of the nuclear pore complex and is anchored there by NDC1. Defects in this gene are a cause of achalasia-addisonianism-alacrima syndrome (AAAS), also called triple-A syndrome or Allgrove syndrome. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of AAAS function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 5' splicing donor site. At least one publication reports experimental evidence that this variant affects mRNA splicing in lymphocytes from a patient carrying homozygous c.1331+1G>A (Kallabi_2016). The variant allele was found at a frequency of 4.8e-05 in 251462 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in AAAS causing Glucocorticoid Deficiency With Achalasia (4.8e-05 vs 0.0011), allowing no conclusion about variant significance. c.1331+1G>A has been reported in the literature in multiple individuals affected with Glucocorticoid Deficiency With Achalasia (example, Kallabi_2016). These data indicate that the variant is very likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 26595337, 27414811).
