SHMT2 (HGNC:10852): (serine hydroxymethyltransferase 2) This gene encodes the mitochondrial form of a pyridoxal phosphate-dependent enzyme that catalyzes the reversible reaction of serine and tetrahydrofolate to glycine and 5,10-methylene tetrahydrofolate. The encoded product is primarily responsible for glycine synthesis. The activity of the encoded protein has been suggested to be the primary source of intracellular glycine. The variant allele was found at a frequency of 2.4e-05 in 251418 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.121_122delAG in individuals affected with Neurodevelopmental Disorder With Cardiomyopathy, Spasticity, And Brain Abnormalities and no experimental evidence demonstrating its impact on protein function have been reported.
