not specified Uncertain:1
The c.46G>A (p.D16N) alteration is located in exon 1 (coding exon 1) of the MYF6 gene. This alteration results from a G to A substitution at nucleotide position 46, causing the aspartic acid (D) at amino acid position 16 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Autosomal dominant centronuclear myopathy Uncertain:1
In summary, this variant is a rare missense change with uncertain impact on protein function. There is no indication that it causes disease, but the available evidence is currently insufficient to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant is present in population databases (rs749523828, ExAC 0.001%) but has not been reported in the literature in individuals with an MYF6-related disease. This sequence change replaces aspartic acid with asparagine at codon 16 of the MYF6 protein (p.Asp16Asn). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and asparagine. -
