Submissions by phenotype
Hypertrophic cardiomyopathy 14 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 1927 of the MYH6 protein (p.Arg1927Leu). This variant is present in population databases (rs730880152, gnomAD 0.008%). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 31983221). ClinVar contains an entry for this variant (Variation ID: 180428). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt MYH6 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

MYH6-related disorder Uncertain:1
The MYH6 c.5780G>T variant is predicted to result in the amino acid substitution p.Arg1927Leu. This variant was reported in an individual with dilated cardiomyopathy (Table S3, Mazzarotto et al. 2020. PubMed ID: 31983221). This variant is reported in 0.012% of alleles in individuals of African descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
Primary familial hypertrophic cardiomyopathy Uncertain:1

Cardiovascular phenotype Uncertain:1
The p.R1927L variant (also known as c.5780G>T), located in coding exon 36 of the MYH6 gene, results from a G to T substitution at nucleotide position 5780. The arginine at codon 1927 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
