ClinVar contains an entry for this variant (Variation ID: 14128). This missense change has been observed in individual(s) with left ventricular noncompaction (PMID: 18506004, 20965760). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 1766 of the MYH7 protein (p.Ala1766Thr). For these reasons, this variant has been classified as Pathogenic. -

This missense variant replaces alanine with threonine at codon 1766 of the MYH7 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with left ventricular noncompaction, which was not present in his unaffected parents (PMID: 18506004, 21551322). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
