The p.I709T variant (also known as c.2126T>C), located in coding exon 1 of the MLH3 gene, results from a T to C substitution at nucleotide position 2126. The isoleucine at codon 709 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

The I709T variant in the MLH3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The I709T variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The I709T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Isoleucine are tolerated across species. In silico analysis predicts this variant likely does not alter the protein structure/function. We interpret I709T as a variant of uncertain significance. -
