Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Jul 10, 2015 | The p.W2371C variant (also known as c.7113G>T), located in coding exon 57 of the FBN1 gene, results from a G to T substitution at nucleotide position 7113. The tryptophan at codon 2371 is replaced by cysteine, an amino acid with highly dissimilar properties. A different amino acid substitution in the same codon, p.W2371R (c.7111T>C), has been described in an individual with a clinical diagnosis of Marfan syndrome based on Ghent criteria (Hung et al. Ann Hum Genet. 2009;73:559-67). This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6494 samples (12988 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. - |
