Variant summary
Our verdict is Pathogenic. Variant got 12 ACMG points: 12P and 0B. PM1PM2PM5PP2PP3_StrongPP5
The NM_000138.5(FBN1):c.4520G>T(p.Gly1507Val) variant causes a missense change involving the alteration of a conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. G1507D) has been classified as Likely pathogenic.

Familial thoracic aortic aneurysm and aortic dissection Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Nov 05, 2019 | The p.G1507V variant (also known as c.4520G>T), located in coding exon 36 of the FBN1 gene, results from a G to T substitution at nucleotide position 4520. The glycine at codon 1507 is replaced by valine, an amino acid with dissimilar properties, and is located in the cbEGF-like #22 domain. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. An alteration at the same amino acid position, p.G1507D, was reported in a cohort of Marfan syndrome patients (Proost D et al. Hum. Mutat., 2015 Aug;36:808-14). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
