This variant is present in population databases (rs200737571, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with HCN4-related conditions. The p.M632I variant (also known as c.1896G>A), located in coding exon 6 of the HCN4 gene, results from a G to A substitution at nucleotide position 1896. The methionine at codon 632 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
