Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00021 in 251454 control chromosomes, predominantly at a frequency of 0.0027 within the East Asian subpopulation in the gnomAD database. This frequency is not significantly higher than expected for a pathogenic variant in BLM causing Bloom Syndrome (0.00021 vs 0.0035), allowing no conclusion about variant significance. c.2839A>G has been reported in the literature as a VUS in settings of multigene panel testing among individuals affected with/undergoing testing for a variety of cancers (example, Kim_2019, Park_2018, Zhang_2019). These report(s) do not provide unequivocal conclusions about association of the variant with Bloom Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
