To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. gnomAD gene constraints suggest that the MAF gene is intolerant of loss of function (pLI = 0.74; https://gnomad.broadinstitute.org/gene/ENSG00000178573?dataset=gnomad_r2_1); however, loss of function loss of function variants, including splicing variants, have not been well documented in individuals with MAF-related disease (Human Gene Mutation Database). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
