The p.S447C variant (also known as c.1340C>G), located in coding exon 9 of the LDLR gene, results from a C to G substitution at nucleotide position 1340. The serine at codon 447 is replaced by cysteine, an amino acid with dissimilar properties. This variant was reported in individual(s) with features consistent with familial hypercholesterolemia (FH) (Kusters DM et al. J Lipid Res, 2013 Sep;54:2543-9; Ten Kate GJ et al. Atherosclerosis, 2013 Apr;227:334-41; Komarova TY et al. BMC Med Genet, 2013 Dec;14:128; Tada H et al. Front Genet, 2022 Apr;13:872056). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
