IL1B (HGNC:5992): (interleukin 1 beta) The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is produced by activated macrophages as a proprotein, which is proteolytically processed to its active form by caspase 1 (CASP1/ICE). This cytokine is an important mediator of the inflammatory response, and is involved in a variety of cellular activities, including cell proliferation, differentiation, and apoptosis. The induction of cyclooxygenase-2 (PTGS2/COX2) by this cytokine in the central nervous system (CNS) is found to contribute to inflammatory pain hypersensitivity. Similarly, IL-1B has been implicated in human osteoarthritis pathogenesis. Patients with severe Coronavirus Disease 2019 (COVID-19) present elevated levels of pro-inflammatory cytokines such as IL-1B in bronchial alveolar lavage fluid samples. The lung damage induced by the Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is to a large extent, a result of the inflammatory response promoted by cytokines such as IL-1B. The variant was identified in dbSNP (ID: rs376289593), LOVD 3.0 and in control databases in 29 of 251464 chromosomes at a frequency of 0.000115 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: South Asian in 24 of 30616 chromosomes (freq: 0.000784), Other in 2 of 6138 chromosomes (freq: 0.000326) and European (non-Finnish) in 3 of 113746 chromosomes (freq: 0.000026), while the variant was not observed in the African, Latino, Ashkenazi Jewish, East Asian, and European (Finnish) populations. The p.Glu10 residue is not conserved in mammals and four out of five computational analyses (SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity.
