Mutations in this gene have been associated with WHIM (warts, hypogammaglobulinemia, infections, and myelokathexis) syndrome. This pathogenic sequence change results in an amino acid frameshift and creates a premature stop codon five amino acids downstream of the mutation, p.Ser339Phefs*6. This pathogenic sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated CXCR4 protein with potentially abnormal function. While this pathogenic sequence change has not previously been described in patients with CXCR4-related disorders, several other truncating variants including a frameshift affecting codon 339 have been reported in multiple affected family members (Hernandez et al., 2003). - |
