These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic. -

MARS2 is a single-exon gene, and there are two truncating variants, c.550C>T (p.Gln184Ter) and c.682_949del (p.Gly228fs), upstream of the next downstream in-frame Met329 having been reported with Combined oxidative phosphorylation defect type 25 and autosomal recessive spastic ataxia-3, respectively (PubMed: 25754315, 22448145). The variant allele was found at a frequency of 2.5e-05 in 241524 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1A>G in individuals affected with Spastic Ataxia 3 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as likely pathogenic. -
