DNA sequence analysis of the BARD1 gene demonstrated a sequence change, c.575C>A, in exon 4 that results in an amino acid change, p.Ser192Tyr. This sequence change does not appear to have been previously described in patients with BARD1-related disorders. This sequence change has been described in the gnomAD database in one individual from the non-Finnish European sub population only (dbSNP rs587782482). The p.Ser192Tyr change affects a moderately conserved amino acid residue of the BARD1 protein. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Ser192Tyr substitution. Due to these contrasting evidences and the lack of functional studies, the clinical significance of the p.Ser192Tyr change remains unknown at this time. -

This missense variant replaces serine with tyrosine at codon 192 of the BARD1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been performed for this variant. In an international breast cancer case-control meta-analysis, this variant was detected in 1/60466 cases and 3/53461 unaffected controls (PMID: 33471991). This variant has been identified in 1/246104 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.S192Y variant (also known as c.575C>A), located in coding exon 4 of the BARD1 gene, results from a C to A substitution at nucleotide position 575. The serine at codon 192 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

ClinVar contains an entry for this variant (Variation ID: 142467). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
