The encoded protein can phosphorylate and activate several proteins involved in the inhibition of DNA replication and mitosis, and can promote DNA repair, recombination, and apoptosis. Studies have shown that this missense change results in skipping of exon 18 and introduces a premature termination codon (PMID: 30199583). The resulting mRNA is expected to undergo nonsense-mediated decay. Experimental studies have shown that this missense change affects ATR function (PMID: 23144622). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 156536). This missense change has been observed in individuals with Seckel syndrome (PMID: 23144622). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces methionine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 1159 of the ATR protein (p.Met1159Ile).
