Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.0005 in 1614018 control chromosomes in the gnomAD database, including 8 homozygotes. The observed variant frequency is approximately 5 fold of the estimated maximal expected allele frequency for a pathogenic variant in SCN5A causing Arrhythmia phenotype (0.0001) (gnomAD database v4). c.5360G>A has been reported in the literature in individuals affected with Arrhythmia (Splawski_2000, Zaklyazminskaya_2013), however it was also found in several healthy controls (e.g. Kapplinger_2010, Ackerman_2004, Ng_2013, Refsgaard_2012). These report(s) do not provide unequivocal conclusions about association of the variant with Cardiomyopathy. One publication reported experimental evidence evaluating an impact on protein function, however does not allow convincing conclusions about variant effect (Hu_2015). The following publications have been ascertained in the context of this evaluation (PMID: 10973849, 15851227, 20129283, 16414944, 11222472, 22378279, 19841300, 22557970, 23861362, 33535892, 25923670).
