The amino acid change results in proline to threonine at codon 627, an amino acid with highly similar properties. In one study, this alteration was predicted to have neutral effect based on 3D structural analysis (Ittisoponpisan S et al. J Endocr Soc, 2018 Aug;2:842-854). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
