This missense change has been observed in individuals with pattern dystrophy and/or retinal dystrophy (PMID: 28041643, 33137351; internal data). ClinVar contains an entry for this variant (Variation ID: 437999). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CTNNA1 protein function. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. The p.S322L variant (also known as c.965C>T), located in coding exon 6 of the CTNNA1 gene, results from a C to T substitution at nucleotide position 965. The serine at codon 322 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear. -
