Submissions by phenotype
not specified Uncertain:1
The Ala1462Thr variant in LAMA4 has been reported in 1 individual with HCM and i n 1 individual with LVH, RV dilation, VT, and CMT (LMM unpublished data) and has not been identified in large population studies. Computational analyses (bioche mical amino acid properties, conservation, AlignGVGD, PolyPhen2, and SIFT) do no t provide strong support for or against an impact to the protein. Additional inf ormation is needed to fully assess the clinical significance of this variant. -
Dilated cardiomyopathy 1JJ Uncertain:1
This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 1462 of the LAMA4 protein (p.Ala1462Thr). This variant is present in population databases (rs727504595, gnomAD 0.02%). This missense change has been observed in individual(s) with left ventricular non-compaction (LVNC) (PMID: 28798025). ClinVar contains an entry for this variant (Variation ID: 178995). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on LAMA4 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Cardiomyopathy Uncertain:1

Cardiovascular phenotype Benign:1
This alteration is classified as likely benign based on a combination of the following: seen in unaffected individuals, population frequency, intact protein function, lack of segregation with disease, co-occurrence, RNA analysis, in silico models, amino acid conservation, lack of disease association in case-control studies, and/or the mechanism of disease or impacted region is inconsistent with a known cause of pathogenicity. -
