This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 42 of the PRKN protein (p.Arg42Pro). This variant is present in population databases (rs368134308, gnomAD 0.006%). This missense change has been observed in individuals with early onset Parkinson's disease (PMID: 11222788, 15584030, 18973254, 27206984). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 644125). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
