Genes affected
TUBB2B (HGNC:30829): (tubulin beta 2B class IIb) The protein encoded by this gene is a beta isoform of tubulin, which binds GTP and is a major component of microtubules. This gene is highly similar to TUBB2A and TUBB2C. Defects in this gene are a cause of asymmetric polymicrogyria. [provided by RefSeq, Mar 2010]

This sequence change replaces cysteine, which is neutral and slightly polar, with serine, which is neutral and polar, at codon 201 of the TUBB2B protein (p.Cys201Ser). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant has not been reported in the literature in individuals affected with TUBB2B-related conditions. ClinVar contains an entry for this variant (Variation ID: 160183). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt TUBB2B protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The c.602G>C (p.C201S) alteration is located in exon 4 (coding exon 4) of the TUBB2B gene. This alteration results from a G to C substitution at nucleotide position 602, causing the cysteine (C) at amino acid position 201 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

The TUBB2B c.602G>C variant is predicted to result in the amino acid substitution p.Cys201Ser. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.013% of alleles in individuals of European (Non-Finnish) descent in gnomAD. Alternative variant at the same codon p.Cys201Phe has been reported de-novo in an individual with tubulin-related dysgyria (Table 1, Stutterd et al. 2021. PubMed ID: 33604570). Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
