The threonine at codon 1522 is replaced by methionine, an amino acid with similar properties. This variant has been reported in one affected individual from a dilated cardiomyopathy (DCM) cohort, as well as in one healthy control (Mazzarotto F et al. Circulation, 2020 02;141:387-398). This variant was also detected in an individual with arrhythmogenic right ventricular cardiomyopathy (ARVC) from a cardiomyopathy genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
