Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with arrhythmogenic right ventricular cardiomyopathy (PMID: 30847666) and in an individual affected with dilated cardiomyopathy (PMID: 31983221). This variant has been identified in 18/281634 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.T1522M variant (also known as c.4565C>T), located in coding exon 23 of the DSP gene, results from a C to T substitution at nucleotide position 4565. The threonine at codon 1522 is replaced by methionine, an amino acid with similar properties. This variant has been reported in one affected individual from a dilated cardiomyopathy (DCM) cohort, as well as in one healthy control (Mazzarotto F et al. Circulation, 2020 02;141:387-398). This variant was also detected in an individual with arrhythmogenic right ventricular cardiomyopathy (ARVC) from a cardiomyopathy genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
