Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251172 control chromosomes. c.3014T>G has been reported in the literature in individuals affected with Cystic Fibrosis along with the well-known pathogenic variant F508del (Dork_1994, Teder_2000, DeBoeck_2005, Salinas_2016). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 15772171, 7525450, 27214204, 10922396). Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
