Actins are highly conserved proteins that are involved in cell motility, structure, integrity, and intercellular signaling. The encoded protein is a major constituent of the contractile apparatus and one of the two nonmuscle cytoskeletal actins that are ubiquitously expressed. Mutations in this gene cause Baraitser-Winter syndrome 1, which is characterized by intellectual disability with a distinctive facial appearance in human patients. A heterozygous missense variant in exon 3 of the ACTB gene that results in the amino acid substitution of Phenylalanine for Leucine at codon 65 (p.Leu65Phe) was detected. The observed variant and a different missense in the same codon (p.Leu65Val) has previously been reported in patients affected with Baraitser-Winter syndrome [PMID: 25052316, ClinVar: VCV000127161.4]. This variant has not been reported in the 1000 genomes, gnomAD (v3.1), gnomdAD (v2.1) and topmed databases. The in silico prediction of the variant are probably damaging by PolyPhen-2 (HumDiv), SIFT and LRT. The reference codon is conserved across species. In summary, the variant meets our criteria to be classified as a likely pathogenic. -
