The c.1816C>T (p.Q606*) alteration, located in exon 14 (coding exon 13) of the RAD21 gene, consists of a C to T substitution at nucleotide position 1816. This changes the amino acid from a glutamine (Q) to a stop codon at amino acid position 606. This alteration is not expected to trigger nonsense-mediated mRNA decay, and impacts the last 4.1% of the protein. Premature stop codons are typically deleterious in nature and a significant portion of the protein is affected (Ambry internal data). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as likely pathogenic. -
