Uncertain significance, criteria provided, single submitter | clinical testing | Women's Health and Genetics/Laboratory Corporation of America, LabCorp | Jun 24, 2022 | Variant summary: CHD7 c.3566G>A (p.Arg1189His) results in a non-conservative amino acid change located in the SNF2, N-terminal domain (IPR000330) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249190 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3566G>A has been reported in the literature in individuals affected with Autism Spectrum Disorder or CHARGE syndrome (Jiang_2013, Butcher_2017, Stessman_2017). These reports do not provide unequivocal conclusions about association of the variant with Hypogonadotropic Hypogonadism 5 With Or Without Anosmia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance - |

CHARGE syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Dec 06, 2022 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CHD7 protein function. ClinVar contains an entry for this variant (Variation ID: 434762). This missense change has been observed in individual(s) with clinical features of CHD7-related conditions (PMID: 23849776, 28191889, 28475860, 29304373). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 1189 of the CHD7 protein (p.Arg1189His). - |
