Variant summary: CHD7 c.3566G>A (p.Arg1189His) results in a non-conservative amino acid change located in the SNF2, N-terminal domain (IPR000330) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249190 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3566G>A has been reported in the literature in individuals affected with Autism Spectrum Disorder or CHARGE syndrome (Jiang_2013, Butcher_2017, Stessman_2017). These reports do not provide unequivocal conclusions about association of the variant with Hypogonadotropic Hypogonadism 5 With Or Without Anosmia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance.
