The glutamic acid at codon 185 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this variant is predicted to be deleterious by in silico analysis. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant has been reported to segregate with disease in one family and in-vitro functional studies showed that it resulted in impaired autophagic function (Gonzalez MA et al.
