The encoded protein plays a role in protein degradation, intracellular membrane fusion, DNA repair and replication, regulation of the cell cycle, and activation of the NF-kappa B pathway. This protein forms a homohexameric complex that interacts with a variety of cofactors and extracts ubiquitinated proteins from lipid membranes or protein complexes. Mutations in this gene cause IBMPFD (inclusion body myopathy with paget disease of bone and frontotemporal dementia), ALS (amyotrophic lateral sclerosis) and Charcot-Marie-Tooth disease in human patients. In addition, this variant is predicted to be deleterious by in silico analysis. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant has been reported to segregate with disease in one family and in-vitro functional studies showed that it resulted in impaired autophagic function (Gonzalez MA et al. Brain, 2014 Nov;137:2897-902). Based on the majority of available evidence to date, this variant is likely to be pathogenic. -

This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 185 of the VCP protein (p.Glu185Lys). -
