Following criteria are met: 0102 - Loss of function is a known mechanism of disease in this gene and is associated with 3-methylglutaconic aciduria, type I (MIM#250950). (I) 0106 - This gene is associated with autosomal recessive disease. (I) 0210 - Splice site variant proven to affect splicing of the transcript with a known effect on protein sequence. Sequencing of patient-derived RNA converted to cDNA, indicates that this variant results in exon 2 skipping. Exon 2 skipping is predicted to result in a shift in the reading frame, and production of a protein expected to undergo nonsense-mediated decay (NMD) (PMID: 16354225).
