The GABA-B receptors inhibit neuronal activity through G protein-coupled second-messenger systems, which regulate the release of neurotransmitters, and the activity of ion channels and adenylyl cyclase. This receptor subunit forms an active heterodimeric complex with GABA-B receptor subunit 1, neither of which is effective on its own. This variant is absent from gnomAD(v3.1.1) suggesting it is not a common benign variant in the populations represented in that database. This variant is absent from ClinVar and to our current knowledge has not been reported in affected individuals in the literature. To our current knowledge, variants in the 3’UTR of GABBR2 have not been reported in affected individuals in the literature. Given the lack of compelling evidence for its pathogenicity, the de novo c.*1906T>C variant identified in the GABBR2 gene is reported as a Variant of Uncertain Significance. - |
