Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with GLA-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 60 of the GLA protein (p.Pro60Thr). The Pro60Thr variant in GLA has not been reported in individuals with cardiomyop athy or in large population studies. Of note, another amino acid change at the s ame position (Pro60Leu) has been reported in 1 individual with Fabry disease (Ve dder 2007). Proline (Pro) at position 60 is conserved in evolution, suggesting t hat a change would impact the protein. Other computational analyses (biochemical amino acid properties, AlignGVGD, PolyPhen2, and SIFT) do not provide strong su pport for or against an impact to the protein. Additional information is needed to fully assess the clinical significance of the Pro60Thr variant. -
